本發明涉及一種酸脅迫抗性提高的畢赤酵母,屬于生物工程技術領域。
背景技術:
在利用有機酸生產菌株,發酵生產有機酸的過程中,隨著有機酸的積累,胞外pH不斷下降,有機酸的解離度也持續下降從而以未解離態進入細胞。細胞質中較高的pH使得有機酸解離,釋放出質子不斷積累,最終導致細胞質酸化并影響細胞代謝的代謝和生長從而影響發酵過程。在工業生產中,通常加入大量NaOH、CaCO3等中和劑以維持培養基的pH穩定。但中和劑的添加也導致了發酵液的滲透壓升高和后期產物純化成本增加。因此提高微生物的有機酸耐受性是提高產量的途徑之一。
畢赤酵母具有表達蛋白易于純化,產量高等優點,因此使用較為廣泛。但是畢赤酵母對酸脅迫的耐受能力較差,不利于研究畢赤酵母作為發酵產酸菌株。
技術實現要素:
為了克服上述問題,本發明提供了一種提高畢赤酵母酸脅迫抗性的方法,所述方法是以Pichia pastoris GS115為宿主、pPICZα為載體表達干酪乳桿菌來源的精氨酰琥珀酸裂解酶ArgH。
在一種實施方式中,所述ArgH的氨基酸序列是SEQ ID NO.1或者SEQ ID NO.2所示的序列。
本發明的第二個目的是提供了一種酸脅迫抗性提高的畢赤酵母基因工程菌,所述畢赤酵母基因工程菌以Pichia pastoris GS115為宿主、pPICZα為載體表達干酪乳桿菌來源的精氨酰琥珀酸裂解酶ArgH。
在一種實施方式中,所述畢赤酵母基因工程菌的構建方法,如下:
(1)通過PCR方法或者化學合成的方法獲得精氨酰琥珀酸裂解酶基因;
(2)將步驟(1)獲得的精氨酰琥珀酸裂解酶基因連接到畢赤酵母表達載體pPICZα上,得到重組質粒pPICZα-ArgH;
(3)將步驟(2)獲得的重組質粒pPICZα-ArgH電轉入Pichiapastoris GS115得到表達精氨酰琥珀酸裂解酶的基因工程菌株。
本發明的有益效果:
(1)本發明通過表達原核來源的精氨酰琥珀酸裂解酶基因,有效提高了真核微生物的酸脅迫抗性。
(2)本發明還優化了精氨酰琥珀酸裂解酶的氨基酸序列,發現優化后的序列能夠更高幅度地提高畢赤酵母的耐酸水平。
具體實施方式
BMGY培養基(g/L):酵母粉10g,胰蛋白胨20g,甘油20g,(NH4)2SO410g,YNB 3.4g,1moL/L KH2PO4-K2HPO4緩沖液(pH6.0)100mL,生物素4×10-5g;
BMMY培養基(g/L):酵母粉10g,胰蛋白胨20g,(NH4)2SO410g,YNB 3.4g,1moL/L KH2PO4-K2HPO4緩沖液(pH6.0)100mL,生物素4×10-5g。
實施例1:重組菌的構建
(1)重組質粒pMD19-T-ArgH的構建
采用化學合成或者PCR的方法,得到含有編碼氨基酸序列為SEQ ID NO.1或者SEQ ID NO.2的基因片段,然后將基因片段連接到pMD19-T質粒上,并轉化大腸桿菌,驗證正確的轉化子,得到重組質粒pMD19-T-ArgH。抽提pMD19-T-ArgH和pPICZα質粒,然后雙酶且pMD19-T-ArgH和pPICZα質粒4h,0.7%瓊脂糖凝膠電泳分析。利用割膠回收試劑盒回收目的片段,16℃連接4h后,利用化學轉化法,轉化大腸桿菌DH5α。篩選正確的轉化子,得到重組質粒pPICZα-ArgH。
(2)畢赤酵母基因工程菌的構建
以SacI線性化的表達載體pPICZα-ArgH電轉化Pichiapastoris GS115。將驗證正確的菌株保種,得到畢赤酵母基因工程菌Pichiapastoris GS115-pPICZα-ArgH。
其中酵母感受態的制備方法如下:
(a)從新鮮YPD平板上挑取畢赤酵母菌落于25mL YPD液體培養基中,30℃,200r/min培養20h;
(b)將0.5mL上述培養液轉接到25mL YPD液體培養基中,30℃,200r/min培養8h(大約OD600=1.3-1.5);
(c)吸取1mL上述菌液于1.5mL無菌的EP管中,4000r/min離心2min,收集菌體,用1.5mL預冷的無菌水,吹打懸浮細胞;
(d)4000r/min離心2min,收集菌體,用1mL預冷的無菌水重新懸浮細胞;
(e)4000r/min離心2min,收集菌體,用1.5mL預冷的1moL/L山梨醇懸浮細胞;
(f)4000r/min離心2min,收集菌體,用100μL預冷的1moL/L山梨醇混勻細胞,5min后方可使用。
電穿孔轉化法如下:
(I)提前預熱電穿孔儀;
(II)取80μL酵母感受態細胞與20μL線性化的表達載體,輕輕混勻,冰浴5min;后轉入預冷的0.2cm電擊杯中;
(III)將電擊杯外水汽徹底擦干凈,放入電擊室;
(IV)按電轉參數:1.5kV,25μF,200,5ms進行電擊;
(V)電擊后立即加入1mL預冷的1moL/L山梨醇于電擊杯中,輕輕吹打后,將內容物全部轉入無菌的1.5mL EP管中,于30℃復活培養1h;
(VI)4000r/min離心5min,去除750μL上清液,將剩余的菌液涂布到含有100μg/mL zeocin的YPDS平板上,30℃培養3-5d。
按照上述方法,得到了2株畢赤酵母基因工程菌Pichiapastoris GS115-pPICZα-ArgH1(表達氨基酸序列如SEQ ID NO.1的酶)、Pichiapastoris GS115-pPICZα-ArgH2(表達氨基酸序列如SEQ ID NO.2的酶)。
實施例2 ArgH蛋白的誘導表達與檢測
(a)將實施例1得到的2株畢赤酵母工程菌分別取單菌落接種于50mL生長培養基BMGY/500mL三角瓶中,30℃,200r/min培養20h;
(b)將上述BMGY生長培養基中的菌液,室溫靜置1h,棄掉上清液,用30mL BMMY培養基重新懸浮菌體,加入100%甲醇至終濃度為1%(v/v),30℃,200r/min培養,每隔24h補加100%甲醇至終濃度為1%(v/v)進行誘導48h。
并用SDS-PAGE蛋白電泳檢測畢赤酵母工程菌中ArgH蛋白得到表達。
實施例3酸脅迫下生長性能試驗
對于考察菌株在脅迫條件下的生長情況,將畢赤酵母基因工程菌Pichia pastoris GS115-pPICZα-ArgH1、Pichia pastoris GS115-pPICZα-ArgH2接種于BMGY生長培養基中的菌液,室溫靜置1h,棄掉上清液,用30mL BMMY培養基重新懸浮菌體,加入100%甲醇至終濃度為1%(v/v),30℃,200r/min培養,誘導12h。
然后將誘導后的菌株接種至新鮮的BMMY培養基中,其中BMMY培養基用乳酸調節至pH 5.0。分別測定Pichiapastoris GS115-pPICZα-ArgH1、Pichiapastoris GS115-pPICZα-ArgH2在脅迫條件下的生長性能,以不表達外源基因的Pichiapastoris GS115-pPICZα為對照。
結果顯示,經生長性能試驗分析,Pichiapastoris GS115-pPICZα-ArgH1菌株相對于Pichia pastoris GS115-pPICZα生物量有約48%的提高,Pichia pastoris GS115-pPICZα-ArgH2菌株相對于Pichiapastoris GS115-pPICZα生物量有約21%的提高,說明在Pichiapastoris GS115中表達了ArgH蛋白后,菌株耐酸性顯著提高。
實施例4酸脅迫條件下耐受性試驗
對于考察菌株對酸的耐受性分析試驗,分別測定了畢赤酵母基因工程菌Pichia pastoris GS115-pPICZα-ArgH1、Pichiapastoris GS115-pPICZα-ArgH2和對照菌株在pH 4.5條件下的存活率。
具體操作方式如下:將菌株誘導培養至OD 2.0,離心收集細胞,經0.85%的生理鹽水洗滌兩次后重懸于等體積的新鮮的pH 4.5(乳酸調節)的BMMY中,脅迫不同時間。脅迫后的菌懸液洗滌兩次后重懸于等體積生理鹽水中,取10μL重懸液,稀釋不同梯度點種于BMMY平板上測定活菌數和存活率。
經耐受性實驗分析,在pH 4.5的BMMY中脅迫6h后,Pichia pastoris GS115-pPICZα-ArgH1、Pichiapastoris GS115-pPICZα-ArgH2的存活率分別是對照菌株的2.56倍、1.49倍,說明畢赤酵母基因工程菌對酸脅迫的耐受性顯著提高。
雖然本發明已以較佳實施例公開如上,但其并非用以限定本發明,任何熟悉此技術的人,在不脫離本發明的精神和范圍內,都可做各種的改動與修飾,因此本發明的保護范圍應該以權利要求書所界定的為準。
